Genome and plasmid sequencing

Our objective is the elucidation of the whole genome sequence of a Lactobacillus buchneri strain. Further, we will sequence versatile Lactobacillus-plasmids, in order to find novel origins of replication. These so called ‘oris’ will be used for the construction of expression and integration vectors.

Today, knowledge of the whole genomic sequence of a microorganism is an indispensable prerequisite for rational genetic engineering. Our target organism,a  L. buchneri protects ensiled forage at aerobic conditions against deterioration and attack by molds by formation of acetate and 1,2-propanediol (Holzer et al., 2003). To elucidate the genetic basis of the involved biochemical pathways and their regulation we have, in cooperation with the CeBiTec in Bielefeld, sequenced the whole genome of this bacterium. Knowledge of the L. buchneri genomic sequence will be used for rational strain improvement.
Additionally we will sequence plasmids obtaind from other LAB in order to identify new origins of replication. These so called ‘oris’ will be used for the construction of novel expression and integration vectors.
